重組IL-1β蛋白(Interleukin-1β,白介素-1β,AbMole,M21094)。實驗人員揭示了乙酰姜酮()通過抑制軟骨細胞鐵死亡改善骨關(guān)節(jié)炎(OA)的作用機制。研究采用體內(nèi)(小鼠內(nèi)側(cè)半月板失穩(wěn)模型,DMM)和體外(IL-1β誘導軟骨細胞OA模型)實驗驗證了:乙酰姜酮可促進軟骨細胞活力、增加細胞外基質(zhì)(ECM)成分的表達、減少金屬蛋白酶(ADAMTS-4、MMP13)和炎癥因子(iNOS、COX2)的釋放,從而抑制軟骨破壞和骨贅形成;乙酰姜酮還通過激活Nrf2/HO-1通路,上調(diào)谷胱甘肽過氧化物酶4(GPX4)的表達,減少脂質(zhì)過氧化產(chǎn)物(MDA)積累,增加谷胱甘肽(GSH)水平,逆轉(zhuǎn)IL-1β誘導的軟骨細胞鐵死亡。
